Reagents SPOLIGO-Biochip for typing of mycobacteria of tuberculosis complex using the specialized microarray (Registration certificate of the Federal Service for Supervision in the Sphere of Health and Social Development № FSR 2012/13826)
The set allows differentiation of Mycobacterium tuberculosis complex, including to distinguish the pathogen of human and bovine type of tuberculosis, as well as to determine the belonging to the main families of Mycobacterium tuberculosis, such as Beijing, LAM9, T1, T5, Haarlem et al., which is important not only from an epidemiological, but also from a clinical point of view. Thus, Beijing family isolates predominate in patients with severe forms of the disease, exhibit high virulence and transmissibility, association with multidrug resistance.
The principle of operation is based on a set of direct repeats the analysis region (DR = direct repeat, English) of the genome of Mycobacterium tuberculosis complex. Investigation of this genetic locus can uniquely determine the strain profile of Mycobacterium tuberculosis complex by the presence / absence of spacers 43 sequences flanked by direct repeats. Methods of analysis using SPOLIGO Biochip-set includes one step PCR and subsequent hybridization of the PCR products on a biochip. The biochip comprises a member 43, each of which contains a unique probe specific to one of the spacers 43 direct repeats region. The interpretation of hybridization results on the biochip through software automatically with delivery of profile information (spoligotypes) strain of Mycobacterium tuberculosis complex in accordance with an international data base SPOLDB4 ( www.pasteur.fr/ip/easysite/pasteur/fr ).
